چکیده مقاله:
Objective: Colorectal cancer is one of the most common gastrointestinal cancers that is second leading cause of death in women after breast cancer and the third leading cause of death in men after lung and prostate carcinoma. The present study aimed to determine the cytotoxic effects of zinc oxide nanoparticles on colon cancer (SW۴۸۰) cell line and measurement of nitric oxide (NO) in cell culture medium.Method: In this experimental study, SW۴۸۰ cells were exposed to ۱۵.۶۲, ۳۱.۲۵, ۶۲.۵, ۱۲۵, ۲۵۰ and ۵۰۰ μg/ml of zinc oxide nanoparticles for ۲۴ hours. Cell viability was evaluated by MTT assay method. NO level was measured in cell culture medium. ANOVA was used to analyze the data.Results: Treatment of SW۴۸۰ cells with zinc oxide nanoparticles resulted in decreasing of viability in a dose dependent pattern. NO concentration significantly increased in response to effective dose of zinc oxide nanoparticles (۱۰۸ μg/ml) compared to control group (P<۰.۰۰۱).Conclusion: The present study suggests that zinc oxide nanoparticles have cytotoxic effects of colon cancer cells. The cytotoxic effect of zinc oxide nanoparticles on colon cancer cells is partly mediated by increasing of NO in cancer cells.
