Introduction: The traditional conception of the mitochondria has been change by discovery of MDPs (mitochondrial-derived peptides). These peptides encode by mitochondrial genome and can translate at the cytoplasm or mitochondria. Humanin (HN) is the first described MDP that has demonstrated cytoprotective effects in various cell types and thus, nowadays it considered as a potential therapeutic target for different cancers and chronic diseases. Method: We searched Humanin, Humanin AND neoplasm, Humanin AND apoptosis in pubmed and scopus data bases to extract articles to investigate and describe the role and the mechanism of Humanin in cancers. Results: The antiapoptotic properties of HN increases the possibility that this peptide can play a role in the pathogenesis of cancer. HN can play a cytoprotective action through inhibiting the mitochondrial or intrinsic apoptotic pathway by modulation of localization and expression of proteins of the Bcl-۲ family. HN can also exert, endocrine, paracrine and autocrine effects by activating or interfering cancer related signaling pathways. For instance, HN triggers signal transducer and activator of transcription ۳ (STAT۳), phosphoinositol ۳-kinases (PI۳K), RAS/mitogen-activated protein kinases (MAPKs), extracellular kinase activation (ERK ۱/۲) and AMPK-eNOS pathways. These findings leads researchers to target HN in different cancers by applying HN antagonists or Local administration of gene therapy vectors that overexpress or silence endogenous HN. Conclusion: Humanin plays a significant role in the pathogenesis of cancers by inhibiting apoptosis and interfering different. It can be a potential therapeutic target for future reaserches.
