miR-۱۲۴۵b-۵p, miR-۹۲a-۳p dysregulation in breast cancer patients and targeted GATA۳

Introduction: microRNAs (miRNAs) are small molecules with stable structure. Recently, from them have been used for diagnostic and therapeutic purposes due to their prominent role in tumor development and metastasis. The current study evaluated the expression of miR-۹۲a- p, miR-۱۲۴۵b-۵p and, their potential target gene, GATA۳ , in breast cancer patients. Material and methods: Thirty-six tumorous and their non-tumorous surrounding tissues from patients with breast cancer were collected. Then, total RNAs were extracted to assess the ۵۱expression of miR-۹۲a-۳p, miR-۱۲۴۵b-۵p, and GATA۳ by qPCR. To evaluate the possible association between miR-۹۲a-۳p, miR-۱۲۴۵b-۵p, and GATA۳, and pothential roles of these miRNAs in regulating EMT and invasion, in silico tools such as miRTarBase, TargetScan, STRING-db, Cytoscape, and miRPath were used. Results: Our findings demonstrated that the expression levels of miR-۹۲a-۳p(-۳.۲۲- fold; P =۰.۰۰۳۱), and miR-۱۲۴۵b-۵p (-۴.۳۵-fold, P =۰.۰۰۳۶) decreased, while the expression level of GATA۳ (+۵.۴۶-fold; P =۰.۰۰۹۷) increased in tumorous tissues in comparison with control tissues. Furthermore, there was a significant association between miR-۹۲a-۳p expression and clinical features, i.e. grade, stage, ER expression, as well as between miR- ۱۲۴۵b-۵p and ER expression and lymph nodes status in corresponding samples. Moreover, meaningful associations between the expression of GATA۳ and grade, stage, tumor size, and ER expression were revealed. Conclusion: miR-۹۲a-۳p and miR-۱۲۴۵b-۵p and their target, GATA۳ , are contributing in breast cancer pathogenesis.The expression and functional these miRNAs and GATA۳ need more study in breast cancer to determine they can be good for targeted therapy.
