Hypericin and Naringenin: Investigating Plausible Synergistic Apoptotic Effect on Y۷۹ Retinoblastoma Cell Line

Retinoblastoma is an eye cancer arises from immature retina cells. According to the detrimental irreparable consequence of the retinoblastoma, finding efficient modality for this cancer is of great importance. According to the anti-cancer impact of hypericin and naringenin, we put the main aim of this study to unravel the apoptotic/anti-cancer effect of these compounds on Y۷۹ retinoblastoma cell line. Methods: By XTT assay, the inhibitory concentration of hypericin for ۲۴ and ۴۸ hours revealed. Cytotoxic effect of naringenin investigated by XTT and trypan blue exclusion assay further confirmed the inhibitory impact of these agents on Y۷۹ cells viability. Flow cytometry Annexin V/PI determined the cell death. The mRNAs expression level of Bax and Bcl-۲ investigated by real-time PCR in different samples. Results: The ۲۴ and ۴۸ hours IC۵۰ of hypericin, calculated to be ۲.۵ and ۱.۲۵ (μg/ml) respectively. ۵۰ (μg/ml) naringenin induced about ۲۰% and ۳۰% apoptosis in Y۷۹ cells after ۲۴ and ۴۸ hours. Trypan blue staining and flow cytometry confirmed this data. Simultaneous treatment with both agents didn’t show synergistic effect. Bax/Bcl-۲ ratio increased in samples treated with hypericin. Hypericin had more cytotoxic effect in Y۷۹ cells compared with naringenin. Furthermore, hypericin and naringenin didn’t have apoptotic synergistic effect in these cells. According to the real-time PCR results, hypericin induces apoptosis in Y۷۹ cells by disrupt the ratio of Bax/Bcl-۲ but narigenin didn’t show significant increase in the Bax mRNA level; although more in vivo/ex vivo experiments needed, according to our results hypericin and naringenin do not show synergistic anti-cancer effect in Y۷۹ retinoblastoma cells. Conclusion: According to the real-time PCR results, hypericin induces apoptosis in Y۷۹ cells by disrupt the ratio of Bax/Bcl-۲ but narigenin didn’t show significant increase in the Bax mRNA level; although more in vivo/ex vivo experiments needed, according to our results hypericin and naringenin do not show synergistic anti-cancer effect in Y۷۹ retinoblastoma cells.
