Hydrolysis Reaction Me chanism of the Second Generation Anticancer D rug Carboplatin: An Ab-inito study

Platinum-based drugs are one of the most important parts of chemotherap y treatments of a variet y of malignancies. The accepted m echanism o f action of cisplatin is double sta ges.۱) intrac ellular activ ation by the hydrolysis of one chloride ligand, drive n by the lower concentration of C l- anions in the cell, ۲) formation o f intra-stran d cross-links in DNA through the covalent bindi ng of the Pt complex t o purine bas es [۱,۲]. De spite the success reached by cisplatin in clinica l application, several serious side-effects and acquired or i ntrinsic resis tance of tum ours are still unsolved problems. In order to ov ercome thes e limitations, a large nu mber of pla tinum compounds such as carboplatin, have been screened in last decades [۳].
