Effects of prolactin on movement disorders and APOE, GFAP, and PRL receptor gene expression following intracerebral hemorrhage in rats

Objective(s): Intracerebral hemorrhage (ICH) occurs mostly in the striatum. In ICH, blood prolactin level increases ۳-fold. The effects of intracerebroventricular injection (ICV) of prolactin on motor disorders will be investigated. Materials and Methods: This study was performed on ۳۲ male Wistar rats in ۴ groups: sham, ICH, and prolactin with ۱ μg/۲ μl (P۱) and ۲ μg/۲ μl (P۲) doses. Results: The weight of animals on days ۱ (P˂۰.۰۱), ۳, and ۷ (P˂۰.۰۵) in the sham and P۲ groups increased compared with the ICH group. Neurological Deficit Score (NDS) in ICH and P۱ groups decreased, and increased compared with sham and ICH groups (P˂۰.۰۰۱), respectively. NDS in the P۱ group increased compared with the P۲ group on days ۱ (P˂۰.۰ ۵), ۳, and ۷ (P˂۰.۰۰۱). The duration time of rotarod in ICH and P۱ groups decreased and increased compared with sham and ICH groups (P˂۰.۰۰۱), respectively. The duration time of rotarod in the P۱ group on days ۳ and ۷ increased compared with the P۲ group (P˂۰.۰۰۱). Travel distance in days ۱(P˂۰.۰۱), ۳(P˂۰.۰۰۱), and ۷(P˂۰.۰۱) decreased in the ICH group. Prolactin receptor (PRL receptor) expression in ICH, P۱, and P۲ groups increased compared with sham and ICH groups (P˂۰.۰۰۱). Glial fibrillary acidic protein (GFAP) expression (P˂۰.۰۰۱) and apolipoprotein E (APOE) (P˂۰.۰۱) expression in the ICH group increased compared with the sham group. GFAP and APOE expression in the P۱ group increased compared with the ICH group (P˂۰.۰۰۱). APOE expression in the P۱ group increased compared with the P۲ group (P˂۰.۰۰۱). Conclusion: According to the results, prolactin reduces movement disorders.
