Evaluation of phenolic and flavonoid compounds in callus from leaf explant of Teucrium polium L. under hormonal condition BAP and IAA

This study aimed to investigate the callus potential and phenolic production of Teucrium polium leaf explant in MS culture medium containing BAP and IAA hormones. Leaf explant was isolated and sterilized from plants grown under hydroponic cultures. The middle cuttings of leaf cultured on media with single and combined hormones BAP (۰, ۰.۵, ۱, and ۱.۵ mg L-۱) and IAA (۰, ۰.۱, ۰.۵, and ۱ mg L-۱) and after eight weeks, the percentage of callus, fresh and dry weight of them as well as phenolic and flavonoid contents were measured. Different levels of hormones had various effects on the callus induction of explant so that the highest percentage of callus (۱۰۰%) related to the combination of "IAA۰.۱ + BAP۱" and "IAA۱ + BAP۱.۵" while the lowest percentage of callus (۷۳%) was in the treatment of "IAA۰.۱ + BAP۱.۵". The highest fresh and dry weights belonged to the callus created on the culture medium with the combined hormone "IAA۱+ BAP۱.۵". The highest content of phenol and flavonoid were obtained in the hormonal treatments of "IAA۱ + BAP۱.۵" Considering there are more than ۶۳% of medicinal plants in our country and in order to prevent the destruction of these valuable and natural plants, it is recommended to research on the potential of callus formation, regeneration, and seedling production in vitro.
