Bacteriophage has been used as strategy for developing new drugs to reduce the incidence of bacterial infections. Here, the bacteriophages were isolated from the sewage water from Semnan University followed by filtering (۰.۴۵μm). The plaques formed using the double-layer agar method. The host range was determined by Spot test. The eclipse period and plaque formation were estimated using the data from the growth curve. Stability at different temperatures and pH values and the optimal MOI was obtained. The plaques were approximately ۱ mm in diameter to infect Pseudomonas aeruginosa. The phage belongs to the family Siphoviridae. Gram-positive strains, Bacillus cereus, and Staphylococcus aureus and gram-negative strains, Pseudomonas aeruginosa were susceptible to the phage. The titer was ۱۰۹ PFU/ml and the eclipse period was ۲۰ minutes, which released ۱۰۰ phage particles from each bacterium. The optimal MOI ۱۰۰ showed the highest stability at ۴ °C and pH ۷. Phage therapy using DDPCC۱۰ phage inhibited Pseudomonas aeruginosa infection
