This study done in the aim of examining the antioxidant potential of aerial parts and leaf callus of Catharantus roseus. In this study, leaf segments of in vitro seedlings were placed on MS medium containing ۲ mg L-۱ ۲,۴-D for induction callus. The phenolic content by Folin–Ciocalteu method and antioxidant potential of ethanolic extracts was measured based on ۱,۱-diphenyl-۲-picrylhydrazyl (DPPH) free radical scavenging, linoleic acid peroxidation inhibition by ferric thiocyanate method (FTC), and malondialdehyde inhibition by thiobarbituric acid method (TBA) against the standards: butylated hydroxytoluene, butylated hydroxyanisole and ascorbic acid. The phenolic content in the aerial parts and callus was ۳۷.۴۲ ± ۱.۱۵ and ۴۸.۳۳ ± ۰.۳۹, respectivly, μg of gallic acid per mg of dried weight extract. In scavenging free radicals (IC۵۰), after standards, aerial parts and callus with concentration of ۲۷۷.۴۵ ± ۴.۴۴۳ and ۳۴۲.۱۱ ± ۲.۶۷ μg ml-۱ , respectively, were able to inhibit half of DPPH• free radicals. After standards, aerial parts and callus extract were able to inhibit ۲۵.۱۷ ± ۰.۹۱ and ۴۰.۳۳ ± ۰.۷۲ percent, respectively, of linoleic acid peroxidation, and in subsequent the inhibit ability of malon dialdehyde was ۳۶.۲۸ ± ۰.۶۸ and ۴۱.۰۲ ± ۰.۵۸ percent, respectively. High correlation was evident between the phenolic content and antioxidant capacity of samples. The results showed that this plant’s callus has phenolic content and antioxidant capacity and this ability can increase with the help of elicitors and while pharmaceutical compounds derived from callus, can use its antioxidant capacity in food and pharmaceutical industries.
