The Effect of oral magnesium sulfate administration on blood SIRT۱ expression in patients with Atherosclerosis

Background and Aim: Atherosclerosis is a chronic inflammatory disease that accounts for a significant percentage of worldwide annual deaths. SIRT۱ has a protective effect against atherosclerosis. SIRT۱ is involved in many cellular processes such as senescence, oxidative stress and inflammation. Magnesium is essential for normal function of endothelial cells and vascular smooth muscle. Although the anti-atherosclerotic effect of SIRT۱ and magnesium is known, the effect of magnesium on SIRT۱ in atherosclerosis is still unknown. The aim of this study was to investigate the effect of magnesium treatment on SIRT۱ expression in atherosclerotic patients.Methods: This study was a randomized, double-blind and placebo-controlled trial. ۴۰ patients with coronary stenosis (۵۰-۷۰%) were selected from cardiology department of Shahid Mohammadi hospital, Bandar Abbas, Iran and randomly divided into two groups of placebo (n=۲۰) and magnesium sulfate treated (n=۲۰). The subject received placebo or magnesium sulfate ۳۰۰ mg/day for ۳ months. The mRNA expression level of SIRT۱ gene was evaluated with Real time PCR at baseline and at the end of treatment.Results: mRNA expression level of SIRT۱ was not significantly different between magnesium treated and placebo groups (p>۰.۰۵).Conclusion: our finding indicated that magnesium sulfate administration at dose of ۳۰۰ mg/day for ۳ months could not affect expression level of SIRT۱. However increase the dose or the time magnesium sulfate treat may increase the expression level of SIRT۱ and could improve the risk factors of Atherosclerosis.
