Extraction and purification of a radish peroxidase for phenolic waste water

Phenolic compounds are one of the major classes of organic pollutants generated through various industrial activities. Among the novel treatment options, an enzymatic process using a peroxidase, such as radish peroxidase, and hydrogen peroxide has proven very effective in removing a variety of phenolic pollutants from aqueous solutions with high efficiency and selectivity. A plant peroxidase extracted from White Radishwaspartially purified by extraction using a polyethylene glycol: ammonium sulphate aqueous two-phase system, followed by gel filtration on a Sephadex G-200 column. Variation of phase composition and NaCl concentration resulted in the desired reduction in volume of the extract and selective partitioning of the enzyme. PEG:ammonium sulphate:NaCl (27:6:2%, w:v) system gave a partition coefficient of 0.18, purification factor of 3.57, volume reduction of 58.75% and bottom phase recovery of 71%. The enzyme was further concentrated by anti-dialysis and was finally purified about 36-fold by gel filtration on a Sephadex G-200 column, with a recovery of 61%. The enzyme was stable at 65°C with an optimum activity at 55°C. The enzyme was stable at pH range 4 - 8 and had an optimum pH at 4.0.
