So far, 23 species of Mycoplasma have been identified and isolated from birds, which poultry and turkeys hosting 16 species but only four of these species are pathogen,including :M.gallisepticum, M.meleagridis, M.iowae, M.synovia.Mycoplasma gallisepticum (MG) is an economically important poultry pathogen since 1954 in Aisa, and causes Mycoplasmosis, that resulting high economic losses through respiratory diseases, genital, urinary tract, eye and joints damages.Objective The purpose of the present studywas to express and purify a chimeric protein Mgc2 –pvpA of Mycoplasma gallisepticum, applicable to diagnostic rELISA test.Methods TRX-mgc2-pvpA-6His was designed andcloned into the Pet 32a+ and transformed into the E. coli BL21 (DE3).The chimeric protein was purified by affinity batch formation method using nickel resin.Results The designed fusion protein was expressed successfully at the optimum incubation temperature 22 ° C for 4 hours and 0.1mM IPTG as an inducer. The recombinant Mgc2 –pvpA protein was expressed in soluble form and high yield of chimeric protein was purified by affinity batch formation method with 38mg / L.Further studies are needed to identify the function of the purified protein and its effects of immune response in the host.
