Corylus avellana cell culture as a supplementary commercial supply of paclitaxel for rare Taxus spp

چکیده

Paclitaxel is an effective antineoplastic agent, which ismainly produced by different species of Taxus. A majorlimitation to the therapeutic use of paclitaxel is thesupply of the compound from its natural source, the barkand needles of low-growth Taxus species. Recently, a fewstudies have introduced hazelnut (Corylus avellana L.)as source of paclitaxel- (and related taxanes). In orderto achieve production of paclitaxel in cell cultures ofhazelnut and compare it with the Taxus, the induction ofcallus formation from seeds of hazel using MS mediumsupplemented by 2,4-dichlorophenoxyacetic acid (2,4-D) and benzyladenine (BA) at different concentrationsand ratios was studied. Suspension cell cultures werealso established in MS medium added with 2,4-D (1mg/L) and BA (0.5 mg/L). Callus induction and cellgrowth rate were measured and growth curve was alsodetermined. Cell-associated and extracellular paclitaxelwere quantified in suspension at 10, 12 and 14 daysafter subculture was determined by HPLC system. Theresults showed that on the growth curve basis, a coupleof week was desired for subculturing in solid and liquidmedia. Over a period of about 6 months, the growth ratein solid and liquid media was about 5 and 15 g/weekrespectively. The content of cell-associated paclitaxel fordays 10, 12 and 14 after inoculation was 1.67, 2.96 and4.25 (μg/g dry weight) respectively. The extracellularpaclitaxel content for above-mentioned days was 12.52,16.16 and 22.25 (μg/L) respectively. If we compare theseresults with those made using Taxus shows that hazelnutin addition to widely available and grows at a much fasterrate than Taxus, its cell cultures also show a greater easeof growth and a similar production in less time, so thatoptimization of the culture medium may provide a goodsystem for this new natural source of paclitaxel
