چکیده
Phosphinothricin (PPT) is the active component of a family of environmentally safe, nonselective herbicides. The bar gene conferring tolerance to the herbicide phosphinothricin (PPT) encodes phosphinothricin acetyltransferase (PAT). Phosphinothricin Acetyl Transferase that detoxifies Phosphinothricin (PPT), the active ingredient of herbicides such as BASTA. We report here a transformation procedure with the bar gene as a selectable marker established via Agrobacterium- mediated transformation. It was introduced into the tobacco genome at a targeted site by homologous recombination. Transgenic plants of Nicotiana tabacum were regenerated by leaf disc transformation using a bar gene as a selectable marker. Transgenic plantlets were selected in medium supplemented with PPT (up to 0.1 mM). The transgenic N. tabacum plants resistant to PPT were positive upon PCR by bar gene specific primers. The transformed plants transferred to a greenhouse proved to be resistant to 1% PPT. The technology demonstrated here could perhaps be usefully transferred to other crop species.کلیدواژه ها
