Introduction & Aim: Hepato Cellular Carcinoma (HCC) represents a major public health problem with a high impact on society and is the third reason of cancer-related deaths worldwide. Because of diagnosis at late stages it has poor prognosis and molecular mechanisms of this cancer has also remained largely unknown. Betatrophin is a novel detected protein which has high level expression in HCC. In this study we examined the effect of this protein on HepG2 cell line. Methods: Recombinant betatrophin was expressed in BL21 and purified by Ni-NTA chromatography. MTT assay was conducted on human liver carcinoma cells (HepG2) to evaluate the cytotoxic effect of recombinant betatrophin on them. Results: It was appeared that betatrophin has low toxicity against HepG2 cancer cells and can reduce cell growth to 30%. Conclusion:It seems that the upregulated expression of betatrophin in hepatocellular carcinoma is more likely to be a response than acting as a tumorgene because otherwise it wouldincrease the proliferation of cells. However additional studies are needed to elucidate the role of betatrophin in HCC.
