چکیده
IntroductionIn recent years, research in the field of cell therapyand tissue engineeringiswidely developed, today there is apotentialfor thepreparation of varioustypes ofscaffoldsfor different cells.ObjectivesThe aim of this work was to elucidate the osteogenesis differentiation of mouse bone marrow-derived (MBMCs) onPLA/CS (Poly lactic acid/ Chitosan) scaffold and in vivo transplantation.MethodsIn this research, we studied surfaceadhesionpropertiesandcell survival of mBMCs on PLA/CS scaffold. Scaffolds were fabricated by an electrospinning technique and cells morphology and cells viability were evaluated by using a scanning electron microscopy (SEM) and MTT assay respectively. mBMCswere then cultivated in the osteogenic conditions on scaffolds for 21 days after which differentiations were evaluated by specific staining as well as qRT-PCR analysis for related gene expression.ResultsThe results of this study demonstrated that the nanfibrous PLA/CS scaffold is an appropriatemodelfor theadhesion, proliferation andcellsurvival of mBMCs cells. Also, in osteoinductive cultures, the cells formed nodules which were positively stained red following alizarin red staining. According to qRT-PCR analysis, the bone-related genes including osteocalcin and osteopontin were expressed.ConclusionThese findings arean important stepin the use ofthree dimensional cultures of stem cells on scaffold in order tocure theosteogenesis diseasesکلیدواژه ها
