Evaluation of nerve growth factor (NGF) methylation status in patients with schizophrenia

چکیده
Nerve growth factor (NGF) is the first and bestcharacterized member of the neurotrophin family. NGF abnormality may be potentially involved in cognitive deficits, such as those observed in schizophrenia. This hypothesis is supported by the finding of decreased plasma levels of NGF in schizophrenia patients compared to healthy controls. Although genetic factors are risk factors for schizophrenia, some environmental factors are thought to be required for the manifestation of the disease.Epigenetic mechanisms regulate gene functions without causing a change in the nucleotide sequence of DNA. It is established that methylation status of the NGF gene is associated with fear learning, memory, and stressful social interactions. This study included 30 patients (20 male and 10 female) with schizophrenia and 40 unrelated healthy controls (20 male and 20 female). Determination of methylation pattern of CpG islands was based on the principle that bisulfite treatment of DNA results in the conversion of unmethylated cytosine residues into uracil, whereas methylated cytosine residues remain unmodified. Methylation-specific PCR was performed with primers specific for either methylated or unmethylated DNA. Statistical analysis was performed using the chi-square test. No substantial difference between patient and control groups was found (p> 0.05). In conclusion, our finding in this research suggests that the hypermethylation of NGF promoter isn t related to schizophrenia formation. However, longer studies with more patient and controls are needed to confirm this result.کلیدواژه ها
