Evaluation of cell adhesion and proliferation of adipose derived mesenchymal stem cells on 3D gelatin scaffold

چکیده
Objective: Extracellular matrix (ECM) by providing a complicated set of biophysical, structural and chemical signals, plays an important role in stem cell adhesion, migration and proliferation. Therefore, recently many researchers have focused on the research of biocompatible scaffolds which provide 3D structure that mimics the natural ECM to improve stem cell proliferation. The aim of this study was to evaluate the cellular behavior of human adipose derived mesenchymal stem cells (hADSCs) on gelatin cryogel scaffold as a 3D culture system and tissue culture polystyrene (TCPS) as a 2D culture system in vitro.At first 3D gelatin scaffolds were fabricated by cryogelation technique and after seeding the hADSCs, the morphology, adhesion and proliferation rate of seeded cells on the scaffolds were assessed using scanning electron microscope (SEM), 3-(4, 5-dimethylthi-azol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assay and 2-(4-Amidinophenyl)-1H-indole-6-carboxamidine (DAPI) staining. HADSCs were cultured in parallel in 2D tissue culture polystyrene and served as control group. Analysis of SEM images and DAPI staining demonstrated that 3D gelatin scaffolds improve adhesion and prolifer-ation rate of hADSCs in comparison to 2D TCPS. MTT assay results showed that fabricated cryogels are biocom-patible and after a defined time period, the number of cells in scaffolds is significantly higher than that of TCPS (P< 0.05).usion: Our findings indicate that 3D gelatin cryogel scaffold can mimic the hADSCs microenvironment and because of highly porous structure provides an extended surface area and therefore improves the hADSCs attachment and proliferation rateکلیدواژه ها
