Cancer is a leading cause of mortality worldwide, despite significant advancements in its diagnosis and treatment. The development of resistance to chemotherapeutic agents, their side effects, and non-specific toxicity necessitate the screening for new anticancer agents. Therefore, the development of new anticancer agents with novel mechanisms of action has become a major scientific challenge. Recently, bacteria and the bioactive compounds produced by them have emerged as a promising alternative for cancer treatment. Bacterial anticancer agents such as antibiotics, bacteriocins, non-ribosomal peptides, polyketides, toxins, and others exhibit various mechanisms of action, including apoptosis, necrosis, reduction of angiogenesis, inhibition of translation and binding, and blocking signaling pathways essential for the elimination of cancer cells. Additionally, targeted live bacteria have provided a unique therapeutic alternative for cancer treatment. In this article, we review the anticancer properties and mechanisms of action of bacterial-derived anticancer agents and tumor-targeting bacteria, along with their potential future applications in cancer therapy.
