Investigation of the Effect of ascorbic acid on proliferative potential of breast cancer cells

چکیده
Breast cancer is one of the most causes of cancer-related mortality in the world. Several factors cause this disease, including genomic instabilities. It is hoped that with control of genomic mechanisms, steps can be taken to improve this disease. Ascorbic acid (AA) is an essential micronutrient that the human body needs. It is also plays biphasic roles in cancers. In this study, we investigated the effects of different doses of ascorbic acid on murine breast cancer cells line (۴T۱). Accordingly, ۴T۱ cells (۳۰۰۰/well) were seeded in ۹۶-well tissue culture plates. After ۴۸ h incubation, we replaced media with fresh media containing increasing doses of AA (۰.۱, ۰.۲, ۰.۵, ۱,۱.۵, ۲, ۲.۵, ۳, ۳.۵ ,۴ ۴.۵ and ۵ mM). After ۲۴ h of incubation, we performed MTT assay by replacing the media with ۱۰ μl of MTT solution and the plates were incubated in the dark for ۳ h. MTT solution was then removed and DMSO was added to the wells. After ۳۰ min extraction at room temperature, the absorbance of the formazan solution was read spectrophotometrically at ۵۷۰ nm. The results of the MTT assay showed that in doses of (۰.۱), (۰.۲) and (۰.۵) mM, the viability rates are more than ۹۰% and in dose of ۱mM ۷۰% cells are alive. The survival of breast cancer cell line (۴T۱) at concentrations higher than ۱ mM was significantly reduced. The results of this study show the sensitivity of breast cancer cells (۴T۱) to high concentrations of ascorbic acid.کلیدواژه ها
