VARICOCELE DISRUPTS THE SERTOLI CELLS AND SPERMATOGONIAL STEM CELLS NETWORK IN THE RIGHT TESTICULAR TISSUE: AN EXPERIMENTAL STUDY

Background and Aim : One of the most common multi-pathophysiology disorders in infertility of men is known as the varicocele (VCL). The VCL is mainly characterized by regraded blood flow into spermatic and cremaster veins in the left portion of testicles. Although, all of the previous investigations showed that the VCL induced left testicular tissue apoptosis that was associated with variuos mechanisms. In the current study we attempted to illustrate the negative impacts of the VCL on the right testicular tissue by evaluating the pathological effects, Sertoli cells and germ cells cross-link and spermatogonial stem cells (SSCs) survival rate.Methods : 24 mature Wistar rats were randomized into control-sham and VCL-induced groups of 6 animals each. Following 2,4 and 6 months the tubular differentiation (TDI), spermiogenesis indices (SPI) and pathology disorders were assayed. The protein levels of GDNF, Gfrα1, and C-Ret were analyzed using immunohistochemistry (IHC) to assess positive cells distribution/one mm2 of tissue. IHC-positive reactions intensity were assessed using the pixel-based frequency analyses by image pro-insight software.Results : Our results revealed that the VCL reduced positive TDI and SPI ratio remarkably versus the control group in a time-dependent manner. In addition, the edema level was significantly enhanced in VCL-induced animals compared to control animals (P<0.05). The VCL-induced animals showed significantly diminished levels of GDNF, Gfrα1, and C-Ret in 4 and 6-month VCL versus the control group (P<0.05). Although GDNF, Gfrα1 and C-Ret protein level reduced in VCL-induced animals, the results in 2-month induced VCL animals showed no significant differences compared to the control group (P> 0.05).Conclusion : It was concluded that the VCL negatively affected left testicular tissue and disrupted the Sertoli cells-SSCs network in the right testicular tissue that resulted in reduction in germ cells self-renewal process.
