Comparing Developmental Competence of mature and immature oocyte in PCOs patients and mRNA levels of AMHR gene as biomarker

Background: In vitro maturation (IVM) could be a good choice for patients who are hormone sensitive and have few oocytes in stimulated cycles. Consequently, selecting embryos with the highest implantation potential is of great importance in assisted reproductive technologies (ART). To date, the choice of the best embryos to transfer is based on morphological parameters. Therefore, movement towards modern technologies such as genomics, transcriptomics, proteomics and metabolomics to select the most competent oocytes and/or embryos with the greatest implantation potential would be significant.Objective: The aim was to assess the developmental competence of the in vitro and in vivo matured human oocytes as well as the AMHR gene expression of cumulus cells (CCs) between tow group in order to evaluate oocyte maturity in PCOs patient.Material and Method: The oocytes and the associated CCs were retrieved from 27 PCO women and divided into groups of GV and MII according to the nuclear maturity status in order to studding developmental competence as well as expression patterns of AMHR gene using real time PCR.Result and conclusion: The fertilization and embryo formation rates were 61.5% and 84.5% vs.67.1% and 90.8% in test and control groups, respectively. There were significant differences in mRNA levels of AMHR gene between the groups. It seems that using immature oocytes could be helpful for patients at risk of ovarian hyperstimulation syndrome (OHSS) as the same as patients with diminished ovarian reserve.
