Anti metastatic effect of Quillaja Saponin on SKOV3 and A2780 ovarian cancer cells

Objective: special metastasis behavior of Ovarian cancer is considered as one of the most important challenges in OC therapeutic methods.Furthermore, the side effect of common ovarian cancer treatment methods have attracted attention into natural product. The aim of the study was to assess the molecular mechanism of Quillaja saponin as natural bio-compound in human A2780 and SKOV3 cancer cells.Methods: A2780 and SKOV3 cells were cultured in RPMI-1640 medium containing 10% FBS and 1% penicillinstreptomycin at 37°C and 5% CO2. cells were treated with different concentrations of Quillaja saponin (20, 30, 40, 50, 80, 100 μg/ml) for 24 hours, then MTT assay was used to calculate 50% inhibitory concentration.The migration of the ovarian cancer cells was evaluated by scratch assay.Results: The results showed that Quillaja Saponin had cytotoxicity against A2780 and SKOV3 cells and inhibited cell proliferation dose-dependently, and IC50 was evaluated as 80 μg/ml .The results of scratch assay after 24 hours demonstrated Quillaja Saponin could prevent invasion of A2780 and SKOV3 cells in a concentration dependent manner.Conclusion: the results showed that Quillaja Saponin reduces the migration of cancer cells.Therefore, Quillaja Saponin could be suggested as an anti-invasive candidate against ovarian cancer.کلیدواژه ها
