چکیده

Background: Sand flies belong to the Phlebotominae , which is one of the six Psychodidae subfamily with more than 800 species identified worldwide, of which 98 species are confirmed and suspected Zoonotic cutaneous leishmaniasis (ZCL) due to Leishmania major is endemic in rural areas of 19 out of 31 provinces of Iran and approximately 80 percent of recorded leishmaniasis cases in the country are ZCL. Phlebotomus papatasi is the main vector of ZCL to human in Iran. Phlebotomus (Paraphlebotomus) caucasicus (Ph. caucasicus) group belongs to the subgenus Paraphlebotomus, which has been considered as a complex species including Ph. caucasicus, Phlebotomus mongoloensis (Ph. mongolensis) and Phlebotomus andrejevi, playing a main role in maintenance of enzootic cycle of L. major among rodent reservoir hosts.Objectives: Determination of infection to leishmania parasites among the wild population of ph. caucasicus and ph. mongolensis in important foci of zoonotic cutaneous leishmaniasis in iranMaterials and Methods: Sand flies were collected from active colonies of rodent burrows from 26 trapping sites using sticky trap paper. In order to detect and identify of Lieshmania parasites in females Ph.caucasicus and Ph. mongolensis, the Nested – PCR amplification of ITS2-rDNA region was performed using the internal and external primers generating amplicon with 245bp for L. major , 206bp for L.gerbilli and 141bp for L. turanica .Results: In the current study we found DNA of different gerbil parasites such as L. major and L. turanica, and mixed infection of L. major /L. turanica in Ph.caucasicus and Ph. mongolensis.Conclusion: Both species Ph. caucasicus and Ph. mongolensis not only participate in the ZCL transmission cycle between reservoir hosts, but also play an important role as secondary vectors in the transmission of leishmaniasis to humans..
