Background: In the different part of world, Canine have a major role in the transmission of viral parasite. Different species of arthropods such as fleas, ticks, mites and luses inhabit on the body of dogs as ectoparasites. Fleas are important ectoparasites and agent of allergic dermatitis. They serve as intermediate host to for parasitic worms, and transmit distinct pathogens. Objectives: The aim of this study is to survey the infection of Dipylidium caninum in fleas of dogs.Materials and Methods: Overall, 947 fleas were collected using brushing against the hair of dogs during (2014-2015). At first fleas were identified morphologically, then they were preserved in 90% ethanol for molecular identification. the 28S ribosomal RNA gene of Dipylidium caninum was amplified using LIN primers in PCR assay. Results: In the morphological study, three species were identified as follows: Ctenocephalides canis, Pulex irritans and Ctenocephalides felis. The results of PCR assay and sequence analysis indicated that 13 Ct. canis specimens on infected dogs were positive for Dipylidium caninum. also, no infection was observed in P. irritans and Ct. felis (Gene bank code: 2077640).Conclusion: According to our observation, Ct. canis was the dominant species circulating species of parasites including Dipylidium caninum among reservoirs
