Preparation and Assessment of Gingerol-loaded Nanoliposomes and their Effect on Breast Cancer Cell Lines (MCF-7)

Cancer is one of the deadliest diseases in the world and breast cancer is the most common cancer amongst women. In recent years, nanocarriers have made a tremendous transformation in the treatment of many diseases. Among these nanocarriers, lipid nanocarriers are of particular importance. Liposome is one of the lipid nanocarriers. Gingerol is an anticancer agent used to treat this disease. However, the use of gingerol has adverse side effects. For this reason, nanoliposomal drug delivery technology was used to reduce the side effects and improve drug efficacy. Certain ratios of phosphatidylcholine, cholesterol and gingerol were combined to prepare nanoliposomal gingrol, the obtained formulation was pegylated using polyethylene glycol 2000 (PEG2000) to improve stability and efficacy as well as solubility. Liposomes were combined with a certain ratio of gingerol in a PBS buffer. Although there are several techniques for liposome preparation, ether injection method was used in the present study. The level of loading gingerol into pegylated liposomal nanoparticle was significantly increased successfully. Then, using a zetasizer, the mean diameters of pegylated nanoliposomal and non-pegylated nanoliposomal gingerol were obtained 111 and 184.5 nm, respectively. The loading efficiency of non-pegylated and pegylated formulation were obtained 92.2 % and 96.5%, respectively. The 48-hour pattern of drug release from nanoliposomes was also evaluated using dialysis method and the results showed that the rates of drug release from non-pegylated and pegylated liposomes in 48 hours were about 4.8% and 3.1%, respectively. This study showed that pegylated nanoliposomal gingerol had a higher cytotoxic effect on breast cancer cell lines as compared with gingerol formulation.
