A study on tomato Gram negative pathogenic bacteria in West Azarbaijan

چکیده مقاله:

The amount of damages caused by tomato bacterial diseases varies in different regions. West Azerbaijan province in terms of weather conditions is suitable for the tomato plants growth. In this province, less tomato fields can be found that lack the symptoms of prokaryotic diseases. In summer 2016, sampling from tomato fields of Urmia, Bukan, Oshnavieh, Khoy, Salmas, Makou, Miandoab and Naghadeh was carried out. Infected tomato plants showed symptoms of leaf necrosis with and without yellow halo, water soaking, wilting and stem soft rotting. Isolation and purification of 215 isolates were done on general media. After performing Gram staining test and selected the Gram negative isolates, the bacteria were evaluated for hypersensitive reaction on tobacco and geranium leaves, and then pathogenicity test was carried out on tomato plants. Phenotypic and biochemical tests including levan on NAS, oxidase, potato soft rot, arginine dihydrolase, obligate aerobic, catalase, starch hydrolysis, Tween 80 hydrolysis, esculin hydrolysis, tolerance of 4, 5 and 7% NaCl, growth at 4 and 40 °C, producing fluorescent pigment on king’B, lecithinase, ketolactose, colony color on NBY and YDC and growth on TTC medium were performed. Results of these experiments indicated that some isolates of genera Pseudomonas, Pectobacterium and Ralstonia are among the studied bacteria. To complete the identification process and determination of the isolates taxonomy, PCR using housekeeping genes is processing.
