The Role of Plant Endophytes in Phytoremediation of Anthracene

چکیده
The relationship between plant and microbes play the main role in the phytoremediation, and it seems that a portion of the ability of plant in removal of PAHs is due to the plant endophytes which degrade PAHs inside the root system. We aim to reveal the presence of gram-positive anthracene degrading bacteria inside the root of ryegrass, which was used for phytoremediation of anthracene. After a greenhouse experiment on phytoremediation of anthracene, the root surface was cleaned and sterilized and then the prokaryotic metagenome was extracted from the roots. The conserve region of the gene encoding naphthalene dioxygenase enzyme which initiates the degradation of anthracene and some other PAHs in gram-positive bacteria were amplified using NMR331f and NMR1134r primers. The results showed that the gram-positive anthracene degrading bacteria were present inside the root system of ryegrass, however there were no amplification in the control plant which were grown in unpolluted soil, indicating that the bacteria from the contaminated-soil penetrate to the root system. The results also showed that anthracene-degrading activity could be accomplished inside the root system of ryegrass which eventually leads to removal of anthracene from soil. It could be concluded that the plant endophytes involve in degradation of anthracene during phytoremediation.کلیدواژه ها
