Study the anticancer effect of herbal Carcumin extract on HER-2 expression in gastric adenocarcinoma cells

چکیده
Introduction &Aim: Gastric cancer (GC) is the most common cancer with high mortality worldwide. Reducing or increasing the expression of genes by identifying their signal pathways can help to increase GC treatment. Her2 is a receptor of tyrosine kinase , which treats cancerous cells like an oncogene. Considering the least side effect of herbal extracts, in this study we have evaluated HER-2 gene expression in AGS cells treated with Carcumin extract. Methods:The AGS cells were incubated 37°C containing 5% CO2 with 85% humidity DMEM with 10% FBS . The cells were treated with concentrations of 800, 1200, 2000 μg /ml of Carcumin for 48 hours. Extraction of RNA, synthesis of cDNA has been done using special kits. The study of HER-2 gene expression was performed by Real time PCR and also GAPDH gene was used as the internal control. Results: The results have shown significant reduction of HER-2 gene expression in all concentrations of Carcumin extract. Conclusion: Carcumin plant is known to have anti- cancer effects and few side effects. It seems that this herbal extract has tremendous anticancer effects.کلیدواژه ها
