The effects of sodium butyrate on the expression of LHX1 gene as a diagnostic biomarker in HCT-116 colorectal cancer cell line

The transcription factor LHX1 is one of the transcription factors involved in the HDAC8 gene. The expression of this factor changed in cancer. Sodium butyrate (NaBt) is a potent growth inhibitor and an agent suppression of gene expression. The aim of this study was to investigate the effects of NaBt as histone deacetylase inhibitors on the expression of LHX1 gene in colorectal cancer cell line. Methods: HCT116 cells were cultured in DMEM and then treated with various concentrations of NaBt (from 6.25mM to 200mM) at 24, 48 and 72 hours. The cytotoxicity was evaluated by MTT assay. IC50 was determined graphically. RNA was extracted from the treated and untreated cells and cDNA was synthesized by yekta tajhiz kit. Quantitative Real-Time-PCR was performed to investigate the gene expression of LHX. Fold change expression of LHX1 gene after treatment was evaluated by (2-ΔΔct) method. Results: Our study revealed that NaBt can inhibit the proliferation of cells via a concentration and time-dependent manner. Moreover, quantitative real- time PCR results showed that in NaBt treatment cells, the expression of LHX1 gene was significantly decreased compared with the control group (untreated cells) (p< 0.05). Conclusion : The current study indicated that NaBt had anticancer activities and inhibit growth HCT-116 human colorectal cancer cell line. Also, NaBt, depending on the concentration and time manner reduced the expression of the LHX1 gene. On the other hand, the LHX1 gene can be used as a diagnostic biomarker for human colorectal cancer.
