Abstract: Highly pathogenic avian influenza (HPAI) is a devasting disease in wild birds and poultry which has spread all over the world and is a serious threat for animal and human health [1,2,3,]. In Iran, in September 2017 virus of the H5N8 subtype was reported in Isfahan and was associated with a vast economic consequence. In this study, the whole genome of the virus and the HA gene compared with other reported cases in the database NCBI were studied and evaluated. Initially, a deep search at the NCBI site in order to find all completely of the virus based on the region found in 2017 and single gene hemagglutinin was accomplished. In the next step, phylogenetic tree construction was conducted by clustalW and MEGA 7, based on both methods K-mer and ML. The results of both trees showed that the strain reported in Isfahan with bootstrap over 80% had a highest genetic similarity to strain Cameroon in Africa. These findings indicate that the virus has entered Iran by migratory waterbirds. This study is helpful for a better understanding of the genetic and antigenic development of H5N8 in the region and the world.
