Modified conditioned medium harvested from MSCs improved cisplatin induced acute renal failure

IntroductionRecently, it is proved that mesenchymal stem cells exert their therapeutic effects through paracrine properties. It is named cell free system . Improvement of this system is considered as new strategy in remedy of diseases including renal failure.ObjectivesTherefore, the present study tries to enrich the conditioned medium with culture of bone marrow stem cells with dimethyl fumarate as an Nrf2 gene inducer.MethodsBMMSCs were cultured with 40μM DMF for 48h. Then the conditioned medium was harvested and stored for further experiments. Acute renal failure was induced in male rats using cisplatin. The blood urea and creatinine were analysed at 24h, 48h and 7 days after cisplatin injection. Conditioned medium was injected intravenously. After 14 days the rats were sacrificed and their renal tissues obtained for histopathological analysis.ResultsOur findings revealed that renal failure successfully induced in the animals according to blood urea and creatinine (P<0.05). Modified conditioned medium improved the renal failure with decrease of blood urea and creatinine. In addition, it seems that enriched conditioned medium restored renal tissue better than control.ConclusionApplication of cell free system is promising in renal failure. Nrf2 induction could induce antioxidants cascade and enriches the harvested conditioned medium. Hopefully, administration of high quality conditioned medium would be a safe and efficient strategy in future
