Study of the proliferation of human mesenchymal stem cells on PLLA nanofibers in the presence of donepezil hydrochloride drug

Mesenchymal stem cells are pluripotent cells which are able to self-renewal and cell differentiation. Donepezil is an acetylcholine esterase inhibitor that use to moderate Alzheimer Disease. Some studies represented the lower risk of hip fracture in donepezil users in comparison with non-users. In this study, we examined the proliferation of human bone marrow derived mesenchymal stem cells (hBMSCs) on PLLA nanofiberous scaffold in the presence of donepezil. Nanofiberous scaffold synthesized by electrospinning technique, then were exposed to oxygen plasma to induce hydrophilicity and better cell adherence. Scanning electron microscopy (SEM) used to study the structure of fabricated PLLA nanofiber and cell adhesion. Four types of culture condition were applied: 1- TCPS, 2- PLLA nanofiber, 3- donepezil and 4- scaffold and donepezil. The adhesion and viability of the cells in the presence of donepezil on the scaffold were evaluated by Acridin Orange staining and MTT assay. MTT results showed that the fabricated PLLA scaffold did not have any cytotoxicity as well as promote the cell attachment and proliferation of hBMSCs cells. Under fluorescence microscope, viable cells appear green color with recognizable plasma membrane whereas, nonviable cells appears a pale orange red color with unclear plasma membrane. Donepezil caused toxicity in more than 50 μ/ml but in the range between 500ng and 1μg it acts as a co-stimulator for cell proliferation and growth. In conclusion, our results indicated that PLLA-donepezil is able to provide a suitable condition to support cell proliferation as well as osteogenic differentiation of stem cell in future.
