A lot of efforts have been dedicated to engineering the delivery of designed materials or drugs due to the great demand of regenerative agents against chronic and non-healing wounds. Although conventional treatment methods have been effective to some extent, the main shortcomings of current approaches are its poor availability, adverse side effects, non-specific targeting and primary developed polymeric systems did not meet the necessities of effective drug delivery systems. Current polymeric systems for sustained delivery of small molecules, growth factors or drugs demand a developed performance from the point of stability, loading efficacy and regenerative effects. Immobilization methods associated with nanomaterial carriers have revealed promise in mimicking the natural wound healing cascade during tissue regeneration by enhancing the delivery of biomolecules.In this review, we focus on the developing delivery systems fabricated by different engineering protocols with emphasis on the architecture of the designed system and its ability to modulate drug delivery as a controlled manner will describe current scientific progresses in the field of regenerative medicine, emphasizing challenges in chronic wounds. Specifically we focus on the newest approaches in direct immobilization and appropriate biomaterials applied for biomolecular loading and delivery. Finally, we deliberate how these next-generations have been used to augment tissue regeneration and discuss upcoming implications for their application in the field of regenerative medicine
