The effect of epothilone B on HB9 expression in motor neuron like cell differentiation of human endometrial stem cells

IntroductionProgress towards replacing motor neurons after spinal cord injury has been achieved by developmental biology to differentiate stem cells to motor neurons. Homeobox 9 (HB9) or also known as motor neuron and pancreas homeobox 1 (MNX1) is a developmental gene which has an essential role in motor neuronal differentiation. Epothilone B (Epo B) is a microtubule stabilization agent that is mentioned to promote axon regeneration after spinal cord injury.ObjectivesThe aim of this study is to evaluate the effect of Epo B on motor neuron differentiation of human endometrial stem cells (hEnSCs).MethodsThe expression of HB9 in motor neuron-like cell differentiation of human endometrial stem cells was evaluated by real-time reverse transcription polymerase chain reaction (RT-PCR) using Rotor-Gene Q-QIAGEN and immunofluorescence (IF). Also, the methylthiazolyldiphenyl-tetrazolium bromide (MTT) test was used for cell viability.ResultsThe results of real-time RT-PCR of differentiation group with Epo B showedsignificant over expression of HB9 compared to the differentiation group without using Epo B. The IF also showed HB9 expression in protein expression level. The MTT test did not show cytotoxicity on hEnSCs.ConclusionTheEop B not only promotes axon regeneration but also has asignificant effect on over expression of HB9 motor neurons
