The absence of immune marker HLA-DR Expression and lack of Rejection in EnSCs derived motor neurons developed with small molecules purmorphamin (PMA)

IntroductionHuman endometrial stem cells (hEnSCs) derived motor neurons are one of the best target for transplantation which are not rejected by the immune system. Cell differentiation using small molecules provide a promising strategy for generating target cells for cell transplantation.ObjectivesWe developed an effective and simple induction protocols to differentiating motor neurons from EnSCs that is useful for transplantation to treat motor neuron disease. Treatments with differentiation medias which contain PMA, promote the development of motor neurons.MethodsReal-time PCR and immunofluorescence (IF) test was done for the expression of neurofilament (NF), acetylcholine transferase (Chat) and nestin. Then flow cytometry was done for the expression of the cell surface antigen human leukocyte antigen – antigen D related (HLA-DR).ResultsIF results showed the expression of NF and Chat. Real-time PCR results demonstrated that the cell treated with PMA expressing neural markers NF and Chat. The flow cytometry results confirmed that EnSCs and cells derived from them are not expressing HLA-DR .ConclusionOn the basis of our finding, in the present research it was stated that motor neurons developed by PMA, do not expressed HLA-DR. Onur finding indicated that EnSCs derived motor neurons developed by inducing small molecules do not stimulate immune system.
