Dual drug loaded PGS/PHB core-shell wound dressing with sequentially releasing ability of anti-bacterial and anti-inflammatory drugs

چکیده

IntroductionBiodegradable wound dressing of poly glycerol sebacate (PGS)/ poly hydroxy butyrate (PHB) was fabricated via the coaxial electrospinning process.MethodsSimvastatin and ciprofloxacin were loaded in the core and shell of the fibers, respectively, of the wound dressing fibers thus prepared. The core/shell fibers were subsequently characterized by fourier transform infrared (FT-IR) spectroscopy as well as the contact angle, swelling, and degradation tests. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) images revealed that the fibers enjoyed a fine and uniform core/shell structure.ResultsResults indicated that, introducing drugs into the polymers would cause fiber diameters to reduce, the dressing samples to become more hydrophilic, and degradation to occur faster. It was also observed that the loaded drugs would deposit separately, one in the core and one in the shell of the fibers, while their release would face no barriers or interventions. Approximately 60% of CIP loaded in the fiber shells was released during the first 24 h, which is ideal for preventing infections at the wound site. SIM deposited in the fiber core, however, exhibited a slower and controlled release behavior, with its release peak recorded after two days. Moreover, the drug-containing samples showed a proper bactericidal activity against both Gram-positive and Gram-negative bacteria.ConclusionIt may be concluded that the drug-laden wound dressing fabricated in this study is capable of releasing the two drugs sequentially and that it is the ideal conditions for controlling infections and reducing wound healing duration.
