Glioblastoma is one of the fatal malignant tumors and achievement to acceptable outcome was not seen because of the biological barrier called blood-brain-barrier. In order to bypass from BBB nanoparticles coated with materials acting as targeting ligand such as poloxamer188 are prosperous way. PLGA(Poly lactic-co-Glycolic acid) is a biocompatible and biodegradable polymer which is a useful carrier for drug delivery systems. Paclitaxel is a chemotherapeutic drug but its clinical usage is limited due to its low solubility in aqueous solvents. The purpose of this study was to prepare PLGA nanoparticles containing paclitaxel covered by outer layer which is PVA and poloxamer188. We prepared nanoparticles by precipitation method. Mean diameter of nanoparticles measured by DLS (Dynamic Light Scattering) and SEM (Scanning Electron Microscopy). Zeta Potential detected by Zeta Sizer. Drug loading, Encapsulation efficiency and in-vitro release profile of drug assayed by UV-vis spectroscopy. Mean diameter, mean zeta potential, drug loading and encapsulation was 241±19 nm, -10.58±0.34 mv, %8.4 and %89, respectively and also drug release extended to 80 h.
