Introduction: Autism Spectrum Disorder (ASD) is one of the developmental disorders, with a multifactorial etiology, starting in childhood. So far, it has been proposed that circadian rhythm and melatonin plays a crucial role in the pathogenesis of ASD, but the issue is open to discussion. Methods: Data collection was performed based on the recent hypothesis about maternal circadian rhythm, melatonin and ASD. Major databases (PubMed, Scopus, Google Scholar, Science Direct, Academic Search Premier and EBSCOHost) were searched, for original studies, review articles and congress abstracts. Results: Both experimental and clinical investigations have found that low maternal levels of melatonin and that’s effects on circadian rhythm can have an adverse effect on development of fetus nervous system. Also, it has been declared that a 6-sulfatoxymelatonin level is meaningfully lower in mothers with an ASD child than in control subjects. Conclusion: Low melatonin levels in mother and disrupted circadian rhythm could be one of the contributors to ASD and possibly intellectual disabilities etiology, but not certain.
