Inhibitory effect of propolis nanoparticles on structural changes of Human Hemoglobin fructation

Diabetes has increasingly prevalence in the world. Glycation and production of advanced glycation end products (AGEs) is the main causes of diabetic complications. Previous results have shown that glycation and fructation has important role in structural and functional changes in Hemoglobin (Hb). Propolis is a resinous hive products which has different biological properties such as antioxidant properties. In attention to the better performance of nanoparticles in treatment, different concentration of propolis nanoparticles (PNP) have been used to investigate the inhibitory effects of it in fructation of human Hb. The results showed that propolis nanoparticles can effectively prevent the formation of AGEs in fructated Hb. The results of UV-Visible spectroscopy and fluorescence emission of heme degradation products indicated that PNP can protect the heme structure and inhibited from production of heme degradation products. In addition, the circular dichorism (CD) results demonstrated that PNP can not induce any significant changes in structural of Hb. In fact, interaction of PNP with fructated Hb effectively prevents the α-helix to β-sheet transitions. The comparing results denote that PNP has better performance than bulk propolis, so that less concentration of PNP have more and completely inhibitory effects in Hb fructation. In conclusion, PNP as a natural product can be considered as an effective drug in treatment of diabetes.
