The rook (Corvus frugilegus) is a bird native to Iran. This bird is omnivorous which indicates great importance ecologically. It is also one of the threatened species on the International union: for the Conservation of Nature s Red List. It is essential to recognize the gastrointestinal tract to understand the physiologic mechanism of pathogens in gastrointestinal tract. The present study examined the different parts of the digestive system and mucosal and submucosa histology, muscular and serous membranes of the rook. In this study, five adult and healthy rooks from both sexes were prepared and transferred to the laboratory. Then, the entire gastrointestinal tract was examined based on the existing protocols. it was cleaned during the stages of clearing, impregnating with paraffin and preparing blocks, cutting into 6 micron thickness pieces, then it was stained through H & E method and studied by optical microscope. The results showed that the esophagus of the rook has mucosal, submucosa, muscular membranes, and also endocrine glands with stratified squamous epithelium and proventriculus has endocrine glands and simple columnar epithelium and also salivary glands. The bird gizzard tissue has a thick, muscular stratum, a layer which is called koilin and mucous glands.
