چکیده مقاله:

Background and Aim: Administration of human adipose-derivedmesenchymal stem cells (ADMSCs) has recently considered as a newtherapeutic approach in clinical trials for widespread indicationsincluding cardiac, neurological, hematological and pulmonary disorders.However, efforts to exploit these cells have been met with only modestsuccess mainly due to the low efficiency of the differentiation process,using animal-derived materials and inadequate expansion of the largequantities of desired cell derivatives under good manufacturing practice.In the present study, we have to access the effects of applying variousnatural small molecules and plant extract on efficiency of differentiatehuman ADMSCs into the definitive mesoderm lineage as the initial stepin differentiation into other mesoderm derivatives, such a cardiomyocyte,hematopoietic, endothelial cell, smooth muscle cell, skeletal musclecell, renal cell, adipocyte, chondrocyte and osteocytes.Methods: For this purpose, first mesenchymal stem cells isolated fromthe human adult fat tissue, propagated and characterized using flowcytometry and relevant markers. In second passage cells transferred to24 well plates and treated with various plant-derived small moleculesmainly Wnt/β-catenin signaling pathway, one of the most conservedintercellular signaling activators including resveratrol, stilbene anddifferent plants and evaluate their effects on their differentiation into thedefinitive mesoderm lineage.Results: Data obtained in this study might lead to formulating a definedsafer and more efficient, cost-effective culture medium for mesodermdifferentiation.Conclusion: These findings can lead to the improvement of the currentapproach toward achieving a reproducible and efficient differentiation,verification, and application of the ADMSCs for clinical purposes.
