چکیده

Background and Aim: Bone tissue engineering aims to regeneratenew functional bone using cells, scaffolds, and clinically applicableosteostimulative factors. Lack of efficient short-term treatment strategiesto stimulate osteogenic differentiation of precursor cells has restrictedthe development of a one-step surgical procedure for bone tissue engineering. Earlier, we developed a cost-effective, patient-friendlyone-step surgical procedure for bone augmentation in patients withbone defects using human adipose stem cells (hASCs). Here we studiedwhether short treatment of hASCs with an osteostimulative factor, fittingwithin the procedural time frame, suffices to induce osteogenesis.Methods: hASCs were shortly (minutes) pre-treated with/withoutosteostimulative factor, seeded on biphasic calcium phosphate, andcultured for 2 weeks in the absence or presence of the osteostimulativefactor. Cell attachment was assessed 30 min after cell seeding. Cellproliferation and osteogenic differentiation were analyzed up to 2 weeks.Results: Our data showed that short osteostimulation enhanced cellattachment to biphasic calcium phosphate compared to non-treatedcells. Compared to untreated controls, short osteostimulation of hASCsincreased proliferation and alkaline phosphatase activity of hASCs.Short osteostimulation of hASCs upregulated the osteogenic markersRUNX2 and SPARC, while this was not observed upon continuousosteostimulation. Short osteostimulation of hASCs also upregulatedVEGF189 expression which may contribute to the promotion ofangiogenesis via stimulation of endothelial cell proliferation.Conclusion: In conclusion, short osteostimulation of hASCs not onlyenhances cell attachment to the scaffold at the time of cell seeding butalso promotes proliferation and osteogenic differentiation of hASCs.This indicates that short osteostimulation for bone tissue regeneration ishighly promising for the treatment of patients with bone defects in a onestepsurgical procedure.
