چکیده

Background and Aim: Tissue engineering is of growing interest inbiomedical research. Mesenchymal stem cells (MSCs) are multipotentstromal cells that can differentiate into the osteocyte, adipocyte,chondrocyte cell type. Recently Scientists have been attracted by stemcells. Three types of stem cells include totipotent, pluripotent, multipotentthat have a capacity of division, self-renewal, and proliferation andalso these cells can differentiation to some cell lines, such as hepatic,kidney and etc. In addition, there have been significant advances in thedevelopment of bone scaffolds with various compositions and threedimensionalconfigurations using a variety of techniques.Methods: The biodegradable poly-L-lactic-acid (PLLA) nanofibershave been widely studied for its applications in tissue engineering andcontrolled release systems because of its good processability, goodbiocompatibility, and suitable mechanical properties. Probiotics as a kindof alive microorganisms that have different types. In the health of human,Bifidobacterium and Lactobacillus have a more effective role than others.Of course, the underlying mechanisms probiotic properties normallydepend on the relationship of probiotics with other microorganisms orto the cross-talk of probiotics with host cells. In the present study, poly-llactic-acid (PLLA) was fabricated by electrospinning and treated with anO2 plasma to enhance surface hydrophilicity, cell attachment and growthpotential.Results: Scaffold characterization was done by scanning electron microscope (SEM). Then, human bone marrow mesenchymal stemcells were pre-condition with nisin and cultured on PLLA nanofibers.Cytotoxicity assay and proliferation of MSCs-nisin were determined byMTT assay and acridine orange staining. MTT assay showed that theproliferation of MSCs-nisin increased on PLLA nanofibers and acridineorange staining confirmed the biocompatibility of PLLA-nisin.Conclusion: In conclusion, it was demonstrated that pre-conditionedMSCs with nisin on PLLA scaffolds could serve as a suitable conditionto support the proliferation of MSCs in vitro for use in bone tissueengineering.
