Background and Aim: Despite the emergence of novel therapeuticadvances regarding MI, ischemic heart disease is conceived as amajor cause of global mortality and morbidity. it made investigators todevelop novel re-vascularization techniques that specifically target themicrovasculature in ischemic myocardium. Several studies have recentlyreported promising results by modifying and enhancing cell-mediatedischemic myocardial repair and regeneration. In this study, we aimed toinvestigate the effect of alginate-gelatin encapsulation on the viabilityand dynamic of rat cardiomyoblasts in vitro.Methods: Rat cardiomyoblasts cell line H9C2 were encapsulated byalginate-gelatin solution and incubated for 7 days, and the control groupwas rat cardiomyoblasts from the same cell line without encapsulation while incubated for 7 days as like the encapsulated group. The level ofextracellular enzymes as like SGPT, SGOT, CPK, and LDH were detectedin supernatants from both groups. The level of intracellular enzymes aslike SOD, GPx, and TAC was measured by biochemical analyses in celllysates. To the examination of cell viability, we used MTT method.Results: We found that encapsulation was able to increase the viabilityof rat cardiomyocytes after 7 days. The increased level of intracellularenzymes including SOD, GPx, and TAC was identified compared to thecontrol cells (P> 0.05). No significant differences were found regardingthe level of SGPT and CPK while the level of LDH and SGOT asextracellular enzymes showed decreasing compared to the control nontreatedcells (P> 0.05).Conclusion: The results of this investigation showed that encapsulationof rat cardiomyoblasts with alginate-gelatin microspheres improves thecell s dynamic and viability.
