EFFECT OF HIGH DOSE VITAMIN D SUPPLEMENTATION IN COMBINATION WITH WEIGHT LOSS DIET ON ANTI-ANGIOGENESIS BIOMARKERS IN OBESE SUBJECTS WITH VITAMIN D DEFICIENCY: A DOUBLE BLIND PLACEBO-CONTROLLED RANDOMIZED CLINICAL TRIAL

Background and Aim: Objectives: Angiogenesis plays an important role in pathogenesis of obesity and recently, targeting the adipose tissue angiogenesis for obesity treatment has become a novel therapeutic approach. In vitro and in vivo studies have demonstrated the anti-angiogenesis effects of vitamin D. This study was aimed to assess the effect of vitamin D supplementation on anti- angiogenesis biomarkers in obese subjects with vitamin D deficiency. Methods: In this double-blind placebo-controlled randomized clinical trial, 44 obese subjects with vitamin D deficiency (25 (OH) vita1min D < 50 nmol/L) were assigned into vitamin D (a weight reduction diet + bolus weekly dose of 50000 IU vitamin D) or placebo group (weight reduction diet + placebo weekly) for 12 weeks. Serum levels of 25(OH) D, Calcium, Parathyroid hormone (PTH), Phosporous, thrombospondin 1 (TSP-1), vascular endothelial growth factor receptor 1 (VEGFR-1,) physical activity and sun exposure were assessed before and after intervention. Results: After 12 weeks, serum 25(OH) D level increased significantly with a simultaneous decrease in PTH in vitamin D group (p< 0.001 for all); changes in serum 25(OH) D, PTH, and TSP-1 were significant between the two groups (p<0.05). There were no significant inter and intra-group changes in calcium, phosporous, and VEGFR-1 concentrations after interventionConclusion: Vitamin D supplementation in combination with weight loss diet increased vitamin D status and TSP-1 levels, with no effect on VEGFR-1 in obese subjects with vitamin D deficiency.
