چکیده

Intoduction: Gastric cancer is one of the most common malignancies and cause of cancer death in the world. DNA methylation plays an important role in development and prognosis of gastric cancer. Hypermethylation of tumor suppressor genes is involved in initiation and progression of many types of cancer. Tumor suppressor gene CYLD negatively regulates various signaling pathways including Wnt, Shh, and Notch. In this study we evaluated promoter methylation pattern of CYLD gene in gastric cancer patients.Materials and methods: Tumoral tissues of thirty gastric cancer patients and their adjacent non-tumoral counterparts were studied. DNA extraction and bisulfite treatment followed by sequencing of PCR products were performed.Results: Promoter hypermethylation of the CYLD gene was found in 14(46%) of the 30 tumoral tissues. Also, 6(20%) hemi-methylated tumor margin tissues were observed.Discussion: CYLD promoter hypermethylation can be associated with gastric cancer risk. Because of DNA methylation is a reversible process, it may be a new target to prevent progression of gastric cancer.
