Hesperetin Decrease Oxidative Stress and Ameliorate Seizures Stage in PTZ-Induced Acute Kindling Model of Epilepsy

چکیده

Epilepsy is a type of neurological disorders that characterized by epileptic seizures. Oxidative stress and reactive oxygen species are considered to play a vital role in the pathogenesis of the disease. Moreover, it can be also consequence of epileptic seizures. In this study, we investigate the effect of Hesperetin (HST) on oxidative stress and epileptic seizures in pentylenetetrazol-induced acute kindling model of epilepsy. Forty-two NMRI mice were randomly assigned to the six experimental groups (G): (G1) Hesperidin 10 mg/kg + PTZ 40 mg/1000; (G2) Hesperidin 20 mg/kg + PTZ40 mg/1000; (G3) Hesperidin 50 mg/kg+ PTZ 40 mg/1000; Hesperidin (gavage) as a pretreatment were administrated in these groups for 7 day and in the last day animal received PTZ (IP); (G4) saline (gavage) for 7 days and in the last day animal received PTZ 40 mg/1000 (IP); (G5) intact group received nothing + PTZ 40 mg/1000(IP); (G6) valproate 300 mg/1000 (IP) 30 minutes before PTZ 40 mg/1000(IP) administration. Behavioral testes carried out and brain tissues homogenates were provided for assessment of oxidative stress markers, including thiobarbituric acid-reactive substances (TBARS) indicator of lipid peroxidation (LPO) and ferric-reducing ability of plasma (FRAP) indicator of total anti-oxidant capacity. Behavioral results of HST administration show marked inhibition of anticonvulsant activity in fully kindled animals. The level of TBARS increased and FRAP decreased in animals which have received HST especially at dose of 10 mg/kg compared to PTZ+Saline receiving group. To conclude, these findings suggest that HST not only enhanced the antioxidant activity, but also facilitated seizures in a mice acute model of epilepsy
