چکیده مقاله:

Introduction: Cancer is the most common cause of mortality and morbidity in world .Thyroid cancer is the most common endocrine malignant tumor and papillary thyroid carcinoma (PTC) is the most common form of thyroid malignancy. The use of paclitaxel as a chemotherapeutic agent has become broadly accepted. The present study aimed to assess the effects paclitaxel have been performed on morphology of papillary thyroid cancer cell line.Materials & Methods: Papillary thyroid cancer cell line (B-CPAP)were transferred to DMEM medium, supplemented with 10% FBS and antibiotics then PTC Cells were seeded in 96-well plates at a concentration of 5 × 103 cells/well and were then incubated for 24 hours, then treated with different concentration of Paclitaxel (0/01 to 100 μg/ml). In order to investigate the anticancer effects of Paclitaxel on morphology of papillary thyroid cancer cell line (B-CPAP), cytotoxicity was examined Ethidium bromide/Acridine orange (EB/AO) staining was used for apoptotic and necrotic cells detection. Result: The results of Ethidium bromide/Acridine orange staining showed that with increasing dose of Paclitaxel, The number of apoptotic cells increased. Comparing the number of apoptotic cells formed after treatment with various concentrations of Paclitaxel, showed significant differences compared with control (P < 0.001). Conclusion: Findings of this study showed that anti-cancer effect of Paclitaxel on morphology of PTC cell line can reduce the number of cells and induces cell death by apoptosis.
