چکیده مقاله:

The Mesenchymal stem cell (MSC) has high potential in regenerative medicine based on their renewal properties and multilinearity differentiation capacity. MSCs have able to differentiate to osteoblasts adipocytes, and cardiomyocyte. These cells have many sources such as bone marrow, umbilical cord blood, amniotic membrane and adipose tissue. Amniotic membrane is a postnatal organ, these cells has not require an invasive method and immune -modulatory properties of MSCs make these cells as the primary choice for allo and xeno transplantation. In this study we Isolated MSC from amniotic membrane and differentiated to cardiomyocytes. In laboratory surface markers were identified with flow cytometry. Then we differentiated to the osteoblast, adipocyte, and studied about their characteristic Then added chang differentiation medium to induce MSC to cardiomyocyte. TGF-β1 and α-SMA were studied with immunofluorescence. After extracting RNA and synthesis CDNA we studied the expression of GATA-4 and C -TNT genes with real time PCR. In conclusion, MCS differentiated to osteoblasts and adipocyte. Studying MCS after treatment showed that these cells morphologically differentiate to cardiomyocyte and the level of expression of mRNA of GATA-4 and C-TNT were higher than the cells without differentiation treatment. study about cardiomyocyte are very important because today there are numerous disease related to cardiomyocyte that they can be lethal, and these genes including GATA-4, C-TNT, α-actinin and connexin have effective role on cardiomyocyte differentiation
